Its random it will appear on a pokemon at random and then if you walk around with that pokemon in you party while others are in you party it will spread. Pokerus only stays on for a few days after the pokemon gets it so if you are going to ev train you have a time limit. Keeping a pokemon in a box with pokerus freezes the time.
