Yes, all offensive moves that deal damage barr Night Shade and Seismic Toss require EVs in the respective stat to deal damage. So Grass Knot follows this rule - it's base power varies depending on the opponent's weight like Pika said, but of course the damage will still increase when EVs are on Special Attack.
