To be brutally honest, I've never really understood Melbourne as a tourist destination. In my mind our point of difference is basically the tram network that that's about it. But there are still some places I'd recommend, some might be more obvious than others:
a. Queen Victoria Market is an easy one, it's the literal embodiment of multiculturalism in this city.
b. Lygon Street in Carlton is really good if you are interested in Italian and European food and culture.
c. Williamstown is a good suburb with some nice views and restaurants and without the high density and business of the CBD, otherwise avoid western suburbs because we're boring until you actually leave Melbourne and go further south. (If you do go down that way though, Queenscliff is a really nice area.)
d. Healesville is so far out of the CBD that I don't ever know if you can call it part of the city, but it's a really nice area and it's nowhere near as busy as the inner city. Healesville Sanctuary is nice, though I don't know how much different it would be from what's offered up in Sydney.
e. Melbourne is sort of famous for some of its graffiti lanes and arcades; they're probably where you'll find whatever might differentiate the city from the rest of the country. Most of the good ones come off Collins Street if I recall correctly. If you're really into that kind of stuff, Bourke Street Mall is a decent option too.
f. I haven't been to Melbourne's museums in years, but they're probably good for visitors if you've never seen the National Gallery of Victoria and such.
Probably forgot a lot, but that's what I can remember off the top of my head.
